Hepadnavirus envelope proteins regulate covalently closed circular DNA amplification.
Primary duck hepatocytes were infected with a mutant duck hepatitis B virus defective in envelope protein but competent for viral DNA synthesis. Cells infected by this mutant accumulated higher levels of viral covalently closed, circular DNA (cccDNA) than those infected by wild-type virus. The accumulation of high levels of cccDNA was due to a failure of the mutant-infected cells to suppress de...
متن کاملHelix-coil Transition in Closed Circular DNA
A simplified model for the closed circular DNA (ccDNA) is proposed to describe some specific features of the helix-coil transition in such molecule. The Hamiltonian of ccDNA is related to the one introduced earlier for the linear DNA. The basic assumption is that the reduced energy of the hydrogen bond is not constant through the transition process but depends effectively on the fraction of alr...
متن کاملDetection of HBV Covalently Closed Circular DNA
Chronic hepatitis B virus (HBV) infection affects approximately 240 million people worldwide and remains a serious public health concern because its complete cure is impossible with current treatments. Covalently closed circular DNA (cccDNA) in the nucleus of infected cells cannot be eliminated by present therapeutics and may result in persistence and relapse. Drug development targeting cccDNA ...
متن کاملThe importance of circular DNA in mammalian gene amplification.
Despite a decade of intensive research, the molecular mech anisms of gene amplification have remained elusive. However, recent molecular analyses and cytogenetic investigations sug gest that one frequently used mechanism for generating this important form of chromosome instability begins with the production of acentric, circular, extrachromosomal DNA mol ecules which replicate autonomously. Gen...
متن کاملThe presence of ribonucleotides in mature closed-circular mitochondrial DNA.
Mitochondrial DNA from mouse and HeLa cells is nicked by alkali and chick-embryo ribonuclease H. It has been concluded that ribonucleotides are present in the closed-circular duplex. A comparative study of the scission rates at pH 13.0 of RNA and of mitochondrial DNAs indicates that there are no more than 10 ribonucleotides in the major fraction of the mitochondrial DNAs.
